چکیده

A vibrational force field for the polypeptide chain has been developed for normal-mode analysis of such molecules. I t can reproduce observed frequencies of known structures to within about 5 cm-l. We review the application of this technique to conformational problems in peptides (@-turns and their model compounds), polypeptides [the aII-helix and crystalline poly(g1ycine II)], and proteins (bacteriorhodopsin and glucagon).

Vibrational Analysis of Crystalline Triglycine*

We have refined vibrational force fields for polypeptides that permit excellent reproduction of the normal mode frequencies of such molecules. This is demonstrated in the present study, in which 80 IR and Raman bands of crystalline triglycine between 1800 and 200 cm-’ are reproduced with an average error of 6 cm-‘. A deuterated sample is shown by normal mode analysis to have remained protonated...

APPLICATION OF TWO-DIMENSIONAL ELECTROPHORESIS AND NIH 3T3 CELL TRANSFECTION ASSAY IN THE STUDY OF TUMOR-AS SOCIATED PROTEINS AND GENOMIC DNA TUMOROGENICITY IN MALIGNANT HUMAN ESOPHAGEAL SPECIMENS

Total protein and DNA extracted from histologically diagnosed normal nonmalignant and esophageal tumor tissues were used for analysis of polypeptides pattern by two-dimensional gel electrophoresis and DNA transforming activity in NIH 3T3 cell transfection assay, respectively. In comparison to normal tissues, eight polypeptides underwent down-regulation or disappeared, while seven polypeptid...

Vibrational analysis of peptides, polypeptides and proteins. XXVII. Structure of gramicidin S from normal mode analyses of low-energy conformations.

Normal mode calculations have been carried out on three low-energy structures of gramicidin S obtained from conformational energy calculations. When the results on the amide modes are compared with observed bands in the infrared and Raman spectra of crystalline gramicidin S and its N-deuterated derivative, one of the structures is clearly disfavored. Of the other two, one is slightly favored, a...

Vibrational optical activity (VOA) is due to the differential response of a molecule to left-circularly polarized (LCP) versus right-circularly polarized (RCP) radiation during a vibrational transition

There are two principal forms of vibrational optical activity (VOA), an IR form referred to as vibrational circular dichroism (VCD) and Raman form known as Raman optical activity (ROA). This paper reports examples of the application of VCD spectroscopy for the determination of the absolute configuration and conformation of chiral molecules, e.g. cyclic -lactams. VCD spectroscopy can be applied...

Raman optical activity of tetra-alanine in the poly(l-proline) II type peptide conformation.

The poly(l-proline) II (PPII) helix is considered to be a major conformation in disordered polypeptides and unfolded proteins in aqueous solution. The PPII conformation can be identified by using Raman optical activity (ROA), which measures the different intensities of right- and left-circularly polarized Raman scattered light from chiral molecules and provides information on stereochemistry as...
